Abstract
Experiments on the fluorescence of acridine orange--DNA complexes at a constant concentration of acridi11e orange show that for a native DNA the highest fluorescence intensity per complexed DNA molecule will occur when the concentration ratio of DNA phosphate groups to acridine orange dye molecules is between 5.5 and 7.2. The corresponding limits for denatured DNA are 6.9 and 8.9. Both absorption and fluorescence studies show that denatured DNA exhibits a greater stacking tendency than does native DNA.
