Abstract
Disc electrophoresis in polyacrylamide gels has been found to be a superior method for the analytical fractionation of histones. The details of the procedure developed for disc electrophoresis are discussed. The use of disc electrophoresis for the quantitative determination of histone subfractions has been found to be feasible. The binding of amidoschwartz stain is a linear function of protein concentration for the three major histone fractions and is independent of the fraction used; measurement of stain in the gels has been by densitometry using a Canalco Model E microdensitometer.
