Abstract

The four parts of this thesis describe studies of the DNA sequence organization of some bacterial plasmids and plasmid-related sequences, and of the structures of some type C RNA tumor viruses. The electron microscope heteroduplex method and related techniques are employed throughout.

In Part I, sequence relations among F13 and related plasmids are established. Fl3 (which contains lac, purE, and proC genes) was formed by a type II excision event. Special F sequences which had been shown to be active in recombination have been identified and mapped within the bacterial portion. of Fl3. Their positions and orientation correlate with the points of origin and direction of transfer of a number of independent Hfr strains. This and other evidence indicate that some Hfr's form by reciprocal recombination between the special sequences located on F and on the bacterial chromosome.
